Improved method for determination of high-density-lipoprotein cholesterol II. Enzymic determination of cholesterol in high-density lipoprotein fractions with a sensitive reagent.

چکیده
A reagent is described for the colorimetric enzymic determination of high-density-lipoprotein (HDL) cholesterol. The reagent can be used with HDL fractions isolated by the various methods of precipitation of low- and very-low-density lipoproteins we investigated. The considerable sensitivity obtained by use of Barham-Trinder's reaction allows the sample/reagent volume ratio to be decreased to 1:80, and major interferences thus eliminated. The response is linear from 100 to 2000 mg of HDL cholesterol per liter. The maximum CV obtained in precision tests was approximately 1% within series and approximately 3% between series. Most of the bilirubin interference is eliminated by adopting a reaction pH of 6.1. Because of its sensitivity, the reagent is particularly suitable for use with HDL fractions isolated after precipitation with polyethylene glycol 6000, which are characterized by a marked dilution. HDL cholesterol determination with the proposed reagent is accurate and precise. Values obtained are in line with those provided for by the Abell-Kendall method. The method can easily be automated.

Improved method for enzymic determination of cholesterol in lipoproteins separated by electrophoresis on thin layer agarose gels.
The cholesterol of lipoproteins, separated electrophoretically on thin layer agarose films, is visualised and quantitated by incubating the gels in an enzymic reagent containing cholesterol esterase and cholesterol dehydrogenase. The individual fractions are quantitated by scanning densitometry. No sample pretreatment is necessary. All major fractions are detected readily. The accuracy of the d...
متن کاملEnzymic determination of cholesterol in serum lipoproteins separated by electrophoresis.
We describe a reliable, straightforward procedure for measuring the cholesterol content of serum lipoproteins separated by electrophoresis on cellulose acetate. The lipoprotein fractions are made visible by use of an enzymic reagent and then quantitated by densitometry. Standard electrophoresis equipment is used and the procedure requires just over 1 h for completion. All major fractions except...
متن کاملComparison of two micromethods for determination of lipoprotein cholesterol in plasma.
We compared the results obtained by a micromethod for the determination of plasma lipoprotein cholesterol, in which electrophoresis is used to separate the lipoprotein fractions (beta-, pre-beta-, and alpha-lipoproteins), with those determinations with ultracentrifugation (low-density, very-low-density, and high-density lipoproteins). Precision of determination (coefficient of variation, CV, %)...
متن کاملAssociation of Lecithin Cholesterol Acyltransferase rs5923 Polymorphism in Iranian Individuals with Extremely Low High-Density Lipoprotein Cholesterol: Tehran Lipid and Glucose Study
Background: The serum concentration of high-density lipoprotein cholesterol (HDL-C) is one of the important heritable risk factors for cardiovascular disease and is a target for therapeutic intervention. In this study, we aimed to evaluate the effects of lecithin cholesterol acyltransferase (LCAT) gene polymorphism rs5923 on LCAT enzyme activity and serum HDL-C concentration. Methods: The study...
متن کاملA study of the use of polyethylene glycol in estimating cholesterol in high-density lipoprotein.
We studied polyethylene glycol 6000 precipitation of lipoproteins other than high-density lipoproteins, before cholesterol is estimated in the supernate. Other lipoproteins in the supernatant fractions were detected by using rocket immunoelectrophoresis. A polyethylene glycol concentration of 75 g/L in the final mixture appeared to be optimal, and results agreed with those obtained by ultracent...
